If we have a JIRA Software and want to add JSD do we have to buy additional plugin licenses?

unlimited JIRA Software users license.

looking at a 250 user (agents) JIRA Service Desk license

Hi Eric.
No really. You would just get the JSD functionality which works as a UI layer on top of your jira software that would act as the backend for your service desk portal but all of your jira plugins would still be there and work as intended. You just need to assign jsd licences for your agents in order for them to get the queues
