We have some people that left our company so we obviously don't expect them to work in JIRA anymore.
I realized we are not able to delete them from JIRA because they have opened or worked on some issues, which makes sense. So what we did is removed their access to JIRA but kept them as-is in JIRA.
Now I would like to know if there is a possibility NOT to assign them any issue or said differently to have their users de-activated. Is this possible (being able to assign issues only to users having a JIRA onDemand access activated?).

Have a look at the permission scheme(s).
These explicitly contain a line like "assignable user" with a list of who can be assigned an issue - it matches a user against groups, specific users, roles and other variable ways of identifying someone (e.g. assignee or reporter)
All you need to do is make sure your "dead" users do NOT match assignable-user any more. A common config is to say "jira-users" = "assignable user", so to fix that, you simply remove dead users from that group.
edit - note that this doesn't affect their accounts, and it won't change issues they are currently assigned to, all it does is remove them from the assignable list.
