I'm setting up an automation rule in Jira that is triggered manually. When configuring the "Get input from users" option, I see that I can set a default value for the input fields.
My question is: Is it possible to use a Jira Smart Value within these default value fields to automatically pre-populate them with information from the current ticket? For example, could I use a smart value to default an input field to the ticket's summary or a custom field's value?

No, that is not possible yet. Here is the open suggestion to add such improvements to the user inputs for the manual trigger, and you may watch / vote for it to see progress:
