Hi there,
From reading other posts I understand that by removing our approved domain under "Products -User Access Settings" we can prevent users from requesting access to our products. Can the same results be acheived by changing the product role from user to none?
Essentially we want all users who need access to make their request via our service desk and then be manually assigned access.
Also, do the settings under "Products -User Access Settings" only apply to new users trying to request access and have no effect on existing user access?

Yes, setting the option to None will prevent users from being able to request access.
You should also look at the User Invites section and change the options there to "Don't allow invites".
The settings do not affect users that already have specific product access.
