First thing: are you asking about the view of the board or for the board and backlog?
Jira boards (and backlogs) are based on a saved filter, and the default is ORDER BY Rank ASC. When that is used, the drag-and-drop features for moving issues up / down in position work.
But if the filter sort order is changed to something else, drag-and-drop no longer works. It cannot because Jira would be unable to calculate the new Rank value. (FYI, there is no separate sort order for the board versus the backlog.)
Let's assume you are only asking about the board. You could use swim lanes queries based on Priority to order first, and then the issues would be by Rank within those swim lanes.

No, that is not possible.
The drag-and-drop (to change issue ranking) only works when the board filter's is only sorted by ORDER BY Rank ASC
As a work-around, you could add quick-filters by priority values and select the ones you are interested in viewing.

A bit late, but you can order your board by priority its just a bit of a pain to do so. Here is what you have to do:
On Jira click the search bar in the top right and without typing anything click enter. You will then see the issue navigator with a search bar and all the project issues in a list.
In the search bar add the following:
-> project = *Project name* ORDER BY priority DESC
-> Once entered, click the save as button above the search bar, give the filter a name and save it.
-> Navigate back to your project.
-> On the sprint board view click the ...
-> Under 'GENERAL' you will see 'FILTER'
-> Click the pencil icon next to the current filter and search for the new filter you just created and hit enter.
-> If you have done everything correct your board will now be ordered by priority set in the card

Please reread the original question and additional posts, as your suggestion does not meet the requested need.
You suggest changing the filter to ORDER BY Priority DESC. But the question asked about ordering by Rank and Priority, while preserving the ability to drag-and-drop for changes in Priority. That is not possible with the existing features of Jira Cloud.
